Abstract: Radiotherapy can be used to treat cancer. The aim is to utilize the therapeutic window to killcancer cells efficiently while sparing surrounding healthy tissue as much as possible. Oneapproach that already reduces complications in vivo is the so-called minibeam radiotherapy.The spatially alternating dose maxima and minima are designed to allow significantly moreeffective tumor control compared to conventional broad dose distribution. This thesis comprisesa collimator study which investigated various collimator models and settings to generate x-rayminibeams of best possible quality. Diverse investigations and adjustments made it possible todevelop an optimal setup. With the help of appropriate dosimetric preparations, two largerin vitro studies with H460 lung cancer cells were conducted. A volume study investigatedwhether cells which received the same dose are damaged differently depending on the amountof medium or DPBS volumes they are covered in. The dose study, on the other hand, aimedto investigate the difference between conventional and minibeam irradiation. A variety ofexperiments have been carried out for both studies.
