Abstract: Developing new effective therapies for Ewing sarcoma (EwS) is crucial to improving patient outcomes, especially in cases of metastatic or recurrent disease. In various stages of drug discovery and especially early preclinical development, cell viability assays are often used to assess the antitumor efficacy of compounds of interest. Here, we review and provide guidelines for the application of resazurin- and ATP-based cell viability assays for drug sensitivity screening in EwS. This includes dose-response drug screening to identify metrics such as potency and to compare sensitivity across multiple cell lines, as well as high-throughput drug screening to identify hit compounds that inhibit EwS cells.
